Alzheimer's disease (AD) and Parkinson's disease (PD) and Amyotrophic lateral sclerosis (ALS, often referred to as "Lou Gehrig's Disease,”) are the most commonly encountered neurodegenerative disorders in the elderly adults. There are 4-5million AD sufferers, 1-1.5 million PD in the Unites States. Approximately 5,600 people in the U.S. are diagnosed with ALS each year. The incidence of ALS is two per 100,000 people, and it is estimated that as many as 30,000 Americans may have the disease at any given time.
AD is characterized by the gradual loss of memory and several other important mental functions. It is the most common cause of dementia in older persons. PD is characterized by slowness of movement, tremor, rigidity and imbalance. ALS is characterized by progressive degeneration of motor neuron cells in the spinal cord and brain, which ultimately results in paralysis and death.
Although significant progress has been made in understanding the pathogenesis of these disorders, a curable therapy is still unavailable so far. The FDA approved AD treatment drugs only have mild effects. Among several FDA-approved drugs for PD treatment, Levodopa is the most effective one for controlling symptoms and is regarded as a gold standard. However, its effectiveness tends to decrease after 4 - 5 years. Some patients may have the so-called "on-off" reactions after taking this drug. There is one FDA approved drug, riluzole, that modestly slows the progression of ALS.
